Controlling cells' environments: A step toward building much-needed tissues and organs
ScienceDaily (Aug. 28, 2011) ? With stem cells so fickle and indecisive that they make Shakespeare's Hamlet pale by comparison, scientists have described an advance in encouraging stem cells to make decisions about their fate. The technology for doing so, reported in Denver at the 242nd National Meeting & Exposition of the American Chemical Society (ACS), is an advance toward using stem cells in "regenerative medicine" -- to grow from scratch organs for transplants and tissues for treating diseases.
"Stem cells have great potential in regenerative medicine, in developing new drugs and in advancing biomedical research," said Laura L. Kiessling, Ph.D., who presented the report. "To exploit that potential, we need two things: first, reproducible methods to grow human stem cells in the laboratory, and second, the ability to make stem cells grow into heart cells, brain cells or whatever kind of cell. Our technology takes a different approach to both of these problems, and the results are very encouraging."
Biologically, so-called pluripotent human embryonic stem cells have not made up their minds about what to become. That's essential because these cells, which are derived from embryos, have the agility to develop into the hundreds of different kinds of cells in a fully-formed human body. But controlling their differentiation has also stood as a major barrier to making the stem cell dream come true and using these all-purpose cells in medicine.
Past approaches to growing and scripting the fate of stem cells have involved adding growth-regulating and other substances to cultures of stem cells growing in the laboratory. These conditions left scientists guessing about exactly what wound up in the stem cells. Kiessling and colleagues are pioneering a new approach that involves using chemically controlled surfaces.
Kiessling previously developed chemically modified plastic and glass surfaces that take much of the guess work out of growing stem cells in laboratory cultures. In the past, scientists grew stem cells on surfaces that contained mouse cells. That left scientists with nagging questions about possible contamination of stem cells with disease-causing animal viruses -- a stumbling block for using stem cells in potential medical applications. And that growth system was what scientists term "undefined." There were variations from batch to batch of mouse cells, and scientists never really knew what the stem cells were coming into contact with and how it might be changing them. The synthetic, chemically-defined, surfaces ended that uncertainty. The approach was inexpensive, simple and a much-needed advance in producing stem cells, Kiessling explained.
With the ability to grow stem cells on the synthetic surfaces under chemically defined, or known, conditions, Kiessling's group took an additional step in their latest research. It found that chemically defined surfaces can exert control over signaling pathways. "Signaling" is how molecules talk to one another and get things done inside a cell. It's how an immune cell knows to fight an infection or how a pancreatic cell determines that more insulin is needed in the bloodstream, for example. By controlling how molecules inside a stem cell communicate, researchers could someday in the future nudge them to become one type of cell or tissue over another.
To see whether a new chemically defined surface could change signaling in a pilot experiment, Kiessling tested cancer cells. The research involved use of a signaling substance, transforming growth factor-beta (TGF-beta), which controls a range of activities, from cell growth to self-destruction.
"The new surfaces give scientists much more control over cells, opening up a wide range of possible future applications," Kiessling explained. Building directly on the results of the pilot study, the surfaces could have applications in wound healing. TGF- beta can help wounds heal, but if it touches healthy skin, inflammation or even a cancerous tumor could develop. "We haven't done this, but you could imagine a bandage that has a localized concentration of the special peptide surface that would recruit TGF-beta just to the wound site," said Kiessling.
The surfaces also could make it easier to manufacture organs and tissues in the laboratory someday. "We think that this strategy, with different sets of peptides (building blocks of proteins) bound to the surface, could direct certain human embryonic stem cells on the surface to become one type of cell and other stem cells to become a second cell type, right next to each other. For the tissue engineering involved in growing replacement organs, you need to organize specialized cells in particular ways like this."

Elder abuse has tragic consequences for the abused, their family and their friends. It's an inbuilt breach of trust which traumatizes not simply the abused but anyone who cares about them.
Due to our societies ? medical advances, folks are living longer than previously yet the modern medical practices that plug and sustain life cannot promise that those saved from death may live in full possession of their mental and physical faculties. A massive portion of the populace today are pensioners living alongside some kind of helped care and it's surmised that by the year 2020 just about sixty million folk over the age of 65 will be living in the U. S.. A significant number of these seniors will need a personal carer or need to live in a long term care facility.
Plenty of the elder alive today require a degree of help that's impossible for their family members to provide, and so they must enroll outside help. Unfortunately, long term care facilities, nursing homes, and hospices already experience major budgetary limits and understaffing that makes a contribution to elder abuse, and if these issues aren't remedied within the next decade, the carelessness and abuse of our pensioners will reach pandemic proportions.
Elder abuse comes in several forms including fraud, theft, sexual mistreatment, neglect and desertion, mental abuse, physical abuse and medical misconduct. It is both difficult to recognise and simple to brush off, as many of its victims don't report the neglect and abuse for fear of further harm, retaliation or due to their own sensations of shame for allowing the treatment to occur. To prevent or halt the abuse of an elder loved one needs learning the alarm signals of elder abuse today.
Physical abuse is the easiest kind of abuse to figure out as there is physical proof of harm. Although the elder injure easily, broken bones, unexplained wounds, marks of pressure or restraint, burns or erosions are all obvious signals of physical abuse.
Elder neglect is another type of abuse and might be indicated by the presence of bedsores, wrong hygiene, unexplained weight loss and enduring medical symptoms not healed by medication.
If an aged loved one exhibits inexplicable character changes, social withdrawal or a unexpected shortage of concentration, this is going to be an indication of oral abuse, victimization or wrong medication.
Eventually, money abuse comes in the form of missing funds or assets and ID theft.

Japan's jobless rate climbs to 4.7 percent in July (AP)
TOKYO ? Japan's jobless rate has risen for the second straight month.
The government announced Tuesday that the seasonally adjusted unemployment rate climbed to 4.7 percent in July, edging up from 4.6 percent in June.
The result reflects the fragility of the world's No. 3 economy, which is wrestling with a strong yen and an increasingly uncertain outlook for the global economy.
But the actual jobs picture may be worse because the data do not include the three prefectures hardest hit by the March 11 earthquake and tsunami. In Iwate, Miyagi and Fukushima prefectures, there are ongoing reports of out-of-work residents unable to find jobs to replace ones that were wiped out by the disaster.

SOUTH WILLIAMSPORT, Pa. (AP) ? An American flag draped around his shoulders, Braydon Salzman couldn't contain his glee when he found California teammate Nick Pratto to give him a postgame hug.
The boys from Huntington Beach are headed home with a Little League World Series championship.
Pratto singled in the winning run with two outs and the bases loaded in the bottom of sixth inning, and Salzman pitched a complete-game three-hitter in a 2-1 victory Sunday over Hamamatsu City, Japan, and the tournament title.
"USA! USA," yelled fans before Pratto's single.
"I was just thinking. 'Oh God, Oh God,' Before I was getting in the box," the 12-year-old Pratto said. "But once I got into the box, I calmed myself by telling myself to just look for a good pitch."
Pratto's clutch hit returned the World Series title to the United States with the type of victory even the big leaguers dream about. A U.S. team has now won six out of the last seven World Series, with Japan's win last year the exception.
Pratto tossed his helmet into the air after rounding first before his teammates mobbed him in the infield. The teams exchanged handshakes at the plate before California's giddy players posed at the mound with their new championship banner.
"My team is physically smaller than most of the teams. We didn't think we would get to this stage," Japan manager Akihiro Suzuki, who fought back tears after the game, said through interpreter Kotaro Omori. "All of the players did such a wonderful job to get to this stage."
With runners on first and second, an error by Japan shortstop Gaishi Iguchi on what could have been an inning-ending double play loaded the bases for California. After a force play at the plate, Pratto smacked a solid liner to center off reliever Kazuto Takakura that brought pinch-runner Eric Anderson home with the winning run.
Pratto did his father, manager Jeff Pratto, proud. Nick Pratto said it was great to have his father as his coach, "but he kind of gets on my nerves sometimes."
It was a fitting end to a tense game marked by excellent pitching and timely defense.
Japan starter Shoto Totsuka struck out five over 4 1-3 innings, giving up a homer to right to California slugger Hagen Danner.
First pitch was delayed more than three hours after the outer bands of Hurricane Irene brought more rain than expected to the Williamsport area.
"The result was bad, but they really tried their best," Suzuki said. "Today's weather was difficult for us to get used too. If the weather was like this in Japan, we wouldn't have played."
The clouds finally started parting midway through the game, and sunshine draped the complex by the time the California players left the stadium to cheers by friends and family.
Neither team could convert on several chances to break open the pitcher's duel earlier in the game.
With runners on first and second in the top of the sixth, third baseman Dylan Palmer blocked the bag from sliding Japan runner Ken Igeta on a bunt play to help get California get out the inning.
California put runners on first and second with two outs in the fifth, but Takakura got a flyout to end the inning.
Playing right field in the second, Takakura also made a running catch on fly down the line to save an extra-base hit with a runner on second.
It was Japan running all over the place in the third, flashing trademark hustle to scratch out the game's first run after Seiya Fujita's solid single to left. Pinch-runner Kaito Suzuki moved to second on a bunt and raced toward third with no one covering. The throw bounced into foul territory, allowing Suzuki to score easily.
In a gracious postgame gesture, Japan's players and coaches lined up and exchanged high-fives with the California kids after Huntington Beach did the traditional victory lap around the stadium warning track.
"It's just a dream come true," Danner said. "I never thought we would be in that spot, let alone winning it."

When a hurricane has you in its sights, we all know we?re supposed to stock up on batteries, candles, water and food. Hmmm?food. If we?re going to be cowering in darkness in our apartments for twelve hours as Irene batters and lashes our streets, that food (and drink) part could be the key to finding the bright side of a potential natural disaster.
But what to pick for those special hurricane snacks? In search of inspiration, we asked some of our favorite Brooklyn food makers, purveyors and chefs what they?ll be stocking up on. And so we present Brooklyn?s ?Hurricane Hoarding Picks: Edition Irene.?
